چکیده

1. A polarographic method of measuring and continuously recording the concentration of oxygen in solution down to the order of 1O-9M has been described. 2. This method has been used to determine the respiration rate of bacteria as a function of oxygen concentration. 3. The results so obtained have shown that the respiration rate of bacteria is related to the oxygen concentration by the Michaelis-Menten equation, and the characteristic Km's of a number of organisms have been determined. 4. The effect of temperature and pH changes on the Km have been investigated. 5. Reasons for the differences in the Km's of different organisms have been discussed and an empirical equation relating Km to the size of the organism has been established.

Crystalline Inorganic Pyrophosphatase Isolated from Baker's Yeast

Crystalline inorganic pyrophosphatase has been isolated from baker's yeast. The crystalline enzyme is a protein of the albumin type with an isoelectric point near pH 4.8. Its molecular weight is of the order of 100,000. It contains about 5 per cent tyrosine and 3.5 per cent tryptophane. It is most stable at pH 6.8. The new crystalline protein acts as a specific catalyst for the hydrolysis of in...

Yeast Inorganic Pyrophosphatase

The magnesium ion-dependent hydrolysis of pyrophosphate catalyzed by yeast inorganic pyrophosphatase is markedly inhibited by calcium ion. The kinetics of inhibition have been measured and analyzed in terms of a model involving strong interactions between CaPPi and two metalcomplexed forms of the enzyme, EMg and ECa. Inhibition constants for these interactions have been estimated by nonlinear r...

Quantification of pyrophosphate in soil solution by pyrophosphatase hydrolysis

A commercial pyrophosphatase from Saccharomyces cerevisiae selectively hydrolyzed sodium pyrophosphate, but showed no significant activity towards a range of other organic and condensed inorganic phosphorus compounds. Pyrophosphate determined by pyrophosphatase hydrolysis accounted for 38 12% (mean standard error of 19 sites) of the non-reactive phosphorus in soil solution obtained by centrifug...

Hydrolysis of Adenosine Triphosphate by Crystalline Yeast Pyrophosphatase

Schlesinger and Coon's report that crystalline yeast inorganic pyrophosphatase, in addition to its known ability to hydrolyze inorganic pyrophosphate in the presence of Mg ions, is also able to catalyze the hydrolysis of ATP and ADP in the presence of Zn ions was confirmed. A systematic study showed that the ratio of 370 of PPase-Mg over ATPase-Zn activities per milligram protein in various pre...

Identification and Characterization of an Ecto-Pyrophosphatase Activity in Intact Epimastigotes of Trypanosoma rangeli

In this study, we performed the molecular and biochemical characterization of an ecto-enzyme present in Trypanosoma rangeli that is involved with the hydrolysis of extracellular inorganic pyrophosphate. PCR analysis identified a putative proton-pyrophosphatase (H(+)-PPase) in the epimastigote forms of T. rangeli. This protein was recognized with Western blot and flow cytometry analysis using an...
